محلول دکلسیفایر
محلول دکلسیفایر محلولی است که دارای اسید هیدروکلریک و اسید فرمیک است و از آن برای دکلسیفیه کردن بافت های دارای ترکیبات معدنی مانند دندان و استخوان  استفاده می شود. اساس انجام عملیات دکلسیفیه کردن بر پایه واکنش شیمیایی بین کلسیم موجود در بافت که معمولا به صورت کربنات، فسفات، اکسالات و اورات و اسید هیدروکلریک و اسید فرمیک است.

Decalcifying Solution
It is necessary to conduct decalcification of bones and other hard tissue for the purpose of microscopic analysis of the sample during regular histological processing. The sample is completely immersed in the decalcifying solution. The length of time needed for demineralization (decalcification) depends on the size and density of the treated sample. Bone and hard tissue decalcification requires inorganic or organic acids, or chelating reagents. Rapid decalcifier solution used for compact bone, based on hydrochloric and formic acid. It acts on all mineralized tissues: compact bone, concretions or calcareous deposits in fleshy tissue. A saline corrector has been added to check tissue swelling caused by acid substances.

Principle:
At the base of the demineralization process is the chemical reaction between the tissue calcium (mainly in the form of carbonate, phosphate, oxalate and urate) and hydrochloric acid and formic acid contained in the decalcifier solution
CaCO3 (insoluble) + 2 HCl = CaCl2 (soluble)+ H2CO3
CaCO3 (insoluble) + HCOOH = Ca(HCOO)2 (soluble) + H2CO3

Fixation technique:
1) Volume ratio specimen/decalcifier: 1 : 100
2) Procedure time 4 - 8 hours - < 5 mm thickness.
3) Post decalcifying procedure: reconditioning of the sample. Rinse with slowly running water for 1 hour or apply 3 changes of PBS pH 7.4, 20 minutes each.
