رنگ آمیزی پاپانیکولائو یا رنگ آمیزی پاپ اسمیر، یک روش رنگ آمیزی چند مرحله ای است که توسط دکتر جرج پاپانیکلائو، پدر علم سیتولوژی، ارتقا یافته است. محلول EA50 یکی از محلول های پاپانیکولائو است که برای رنگ آمیزی لام های سیتولوژی و پاتولوژی مورد استفاده قرار می گیرد. این رنگ آمیزی جهت تشخیص انواع سرطان ها مانند سرطان رحم، سرطان دهانه رحم و … استفاده می شود. همچنین از این محلول ها برای رنگ آمیزی انواع ترشحات و سلول ها نیز استفاده می گردد.

Papanicolaou’s solution EA 50
The cytological staining technique developed by Papanicolaou is still the standard method in cancer and hormone cycle diagnosis.

Diagnostic Implications
For routine diagnostic cytology to aid in the identification and classification of exfoliative cells.

Application:
Cytoplasmic staining solution for Papanicolaou method.
For the execution of the staining method is required the use of reagents Papanicolaou Harris hematoxylin and Papanicolaou OG6. Papanicolaou stain imparts a characteristic range of coloration to exfoliative cells of vaginal, cervical, prostatic and other body secretions, allowing critical examination of nuclei and cytoplasmic components.

Principle:
A highly selective blue nuclear stain, Harris’ hematoxylin, is combined with EA50 polychromic mixture, a subtle cytoplasmic stain which differentiates cyanophil cells from eosinophil ones. The last ingredient is OG6 solution, which stains keratinized elements.

Method:
1) Ethanol 95° 2 minutes
2) Distilled water 2 minutes
3) Harris Hematoxylin 1 minute
4) Tap water 5 minutes
5) Ethanol 95° 15 seconds
6) OG 6 2 minutes
7) Ethanol 95° 15 seconds (twice)
8) EA 50 5 minutes
9) Ethanol 95° 15 seconds
10) Absolute Ethanol 30 seconds (twice)
11) Xilene or Bio Clear 2 minutes (twice)

Results:
